    Getting therapeutic drugs into the human brain is not easy. It's a well protected structure, not only because it is encased in bone, but also because blood vessels that serve the brain are lined by tightly- packed cells that make up the 'blood-brain barrier'. That security is there to protect brain tissue from dangerous pathogens and foreign molecules, but it's also a barrier to the entry of therapeutic agents. However, there could be a way to sneak drugs past those barriers by opening them transiently, explains Dr Matthew Campbell from Trinity College Dublin. He's part of a team pioneering an approach that uses specifically designed small interfering RNA to suppress production of a protein called claudin-5, a key component of the barrier that stops drugs moving across from the bloodstream. Injecting siRNA systemically has been shown to improve the permeability of blood cells around brain tumours called gliobastomas in pre-clinical models. "Given the life-threatening nature of this sort of condition, systemic administration of siRNA targeting claudin-5 to allow periodic drug access to tumours would be ethically justifiable," says Dr Campbell. The approach could also be used in the treatment of traumatic brain injury, where suppressing claudin with siRNA could open the blood-brain barrier and encourage the movement of water away from the injury site, thereby reducing the potentially lethal effects of swelling, he added. In a related project, the team has also developed an inducible system to suppress claudin-5 and open the barriers in blood vessels serving the retina. That offers options to treat degenerative eye diseases with systemic drugs rather than repeated injections into the eye.

    Cancer is a complex disease but looking for useful 'biomarkers' in samples from individual patients can ultimately help tailor and improve their treatment. Manually screening slides of tissue sections for such markers is time - consuming and open to variability, but computational methods can help to quantify markers quickly in large numbers of samples, explains William Gallagher, Chief Scientific Officer of OncoMark Ltd. and associate professor of cancer biology at UCD. He and his research team have developed an imageanalysis platform -- IHC-Mark -- that can recognise the features of tumour cells and quantify them in highthroughput screening of samples.The computational approach grew out of a need in Gallagher's research team at OncoMark and the Conway Institute: "We were using tissue micro-array technology where we put hundreds of clinical specimens from cancer patients on to one pathology slide, and you would have a problem analysing that manually." So they developed the system to analyse samples and intelligently quantify markers, and the resulting technology has now been benchmarked against pathologist testing for multiple markers in tissue samples from breast, lung, bladder, melanoma and ovarian cancer patients.

    Business Process Management Software (BPM) is big business, worth $3 billion a year, according to Gartner. Despite this, and the emergence of a market for Automated Business Process Discovery (ABPD) software, there are no tools that automatically discover enduser processes. Yet, this is where most staff conduct business —working with spreadsheets and other applications in uncontrolled and unmapped processes. Which is why a team at Dundalk Institute of Technology (DkIT) have developed a way to improve the visibility of business processes that allows managers to track activities to help increase productivity and cost effectiveness. The technology focuses on spreadsheets. Why target spreadsheets? "Spreadsheets are corporate IT glue, they are used for critical business flows and to fill gaps between systems. So, spreadsheet activity can tell you a lot about how people conduct business and interact with systems" explains Dr Rónán Mac Ruairí, a lecturer and researcher at DkIT. Seeing room for improvement, the team developed Luminous, a technology to allow companies to get a detailed and continuous view of spreadsheet usage. "It's all about visibility - if you can't see a process you can't manage it," says Dr Mac Ruairí. "The system works through a series of software 'sensors' deployed over a network, or on individual workstations, to build up a map of how resources are being used." "Luminous has been undergoing live trials with several companies and has already highlighted areas where processes could be improved and productivity gains made. The technology is of particular use when assessing resource usage before buying or upgrading enterprise resource planning (ERP) systems, and can improve return on investment" he adds.

    The volume of data carried in optical communication networks is increasing due to the increased use of video applications and the surge in cloud computing. Datacentres have huge volumes of data flowing internally and externally in fibres. Fibre to the home is imminent in several countries. Metropolitan areas and backbone already use fibre but need to accommodate more data in each fibre. There is a clear need to put more data through each fibre (i.e. improving its spectral efficiency) rather than installing more fibre. Methods proposed include advanced modulation techniques, multiple wavelengths in each fibre and dynamic reconfiguration of the network using tunable lasers to assign capacity where most needed. Researchers from the Radio and Optical Communications Laboratory in DCU under the guidance of Prof. Liam Barry and Dr. Philip Perry have developed a unique capability to switch and re-route spectrally efficient data paths carrying multiple streams of 10 Gbps data in less than 100 nanoseconds. Pilot Photonics, a proposed spin-out from this group, will produce optical transceiver products that will enable the development of fast switching, high capacity, low energy nodes that will form the fabric of next generation fibre optic communication.

    Getting therapeutic drugs into the human brain is not easy. It's a well protected structure, not only because it is encased in bone, but also because blood vessels that serve the brain are lined by tightly- packed cells that make up the 'blood-brain barrier'. That security is there to protect brain tissue from dangerous pathogens and foreign molecules, but it's also a barrier to the entry of therapeutic agents. However, there could be a way to sneak drugs past those barriers by opening them transiently, explains Dr Matthew Campbell from Trinity College Dublin. He's part of a team pioneering an approach that uses specifically designed small interfering RNA to suppress production of a protein called claudin-5, a key component of the barrier that stops drugs moving across from the bloodstream. Injecting siRNA systemically has been shown to improve the permeability of blood cells around brain tumours called gliobastomas in pre-clinical models. "Given the life-threatening nature of this sort of condition, systemic administration of siRNA targeting claudin-5 to allow periodic drug access to tumours would be ethically justifiable," says Dr Campbell. The approach could also be used in the treatment of traumatic brain injury, where suppressing claudin with siRNA could open the blood-brain barrier and encourage the movement of water away from the injury site, thereby reducing the potentially lethal effects of swelling, he added. In a related project, the team has also developed an inducible system to suppress claudin-5 and open the barriers in blood vessels serving the retina. That offers options to treat degenerative eye diseases with systemic drugs rather than repeated injections into the eye.

Manually screening slides of tissue sections for such markers is time - consuming and open to variability, but computational methods can help to quantify markers quickly in large numbers of samples, explains William Gallagher, Chief Scientific Officer of OncoMark Ltd. and associate professor of cancer biology at UCD. He and his research team have developed an imageanalysis platform -- IHC-Mark -- that can recognise the features of tumour cells and quantify them in highthroughput screening of samples.The computational approach grew out of a need in Gallagher's research team at OncoMark and the Conway Institute: "We were using tissue micro-array technology where we put hundreds of clinical specimens from cancer patients on to one pathology slide, and you would have a problem analysing that manually." So they developed the system to analyse samples and intelligently quantify markers, and the resulting technology has now been benchmarked against pathologist testing for multiple markers in tissue samples from breast, lung, bladder, melanoma and ovarian cancer patients.
